Considering the delicate nature of a microscope, it should be handled with proper care. When moving microscope, it should be grasped by its arm supporting its weight with the other arm on the base. A microscope makes things which cannot be viewed with naked eyes visible for instance bacteria. Nevertheless, if you do not know how to focus on the object, you might not see the object as it requires setting up the microscope properly. In this paper, we are going to examine how to focus a microscope.

The focusing process entails modifying the relationship between the microscope optical system and the examined object to obtain image visibility. During focusing the head is lowered on one side till the eyes are levelled to the microscope stage. T he coarse adjustment knob is constantly used to lower the body and help in the positioning of the lens. The revolving nosepiece positions the high-power lens and focuses it on the object. The fine adjustment knob carries out the final step in focusing to bring the object into precise focus.

Adjusting the microscope from lower to a higher power increases specimen magnification. The image magnified is equal to the eyepiece magnification, multiplied by objective lens magnification. When the magnification subject is lowered, the depth of field increases. As for the subject magnification, when the aperture value increases or the aperture diameter is made smaller, it increases the DOF, and the reverse occurs when you the aperture lens value decreases, or the aperture diameter becomes larger. DOF represents the measurement of the thickness of the focus plane. It decreases with the increase in magnification. The working distance is situated between the specimen and objective lens which decreases with an increase in magnification. The high-power objective lens is required to be nearer to the specimen than the low-power objective lens to keep a sharp focus.

The microscope has many uses in the scientific field, as a biological Scientists, I would use it in viewing microscopic organisms and in identifying pathogens in tissue samples. The microscopes are used in research to examine the behaviour of small organisms that are too very tiny to be view with bare eyes, such as bacteria.
