Adjusting the microscope from lower to a higher power increases specimen magnification. The image magnified is equal to the eyepiece magnification, multiplied by objective lens magnification. When the magnification subject is lowered, the depth of field increases. As for the subject magnification, when the aperture value increases or the aperture diameter is made smaller, it increases the DOF, and the reverse occurs when you the aperture lens value decreases, or the aperture diameter becomes larger. DOF represents the measurement of the thickness of the focus plane. It decreases with the increase in magnification. The working distance is situated between the specimen and objective lens which decreases with an increase in magnification. The high-power objective lens is required to be nearer to the specimen than the low-power objective lens to keep a sharp focus.
